Semaglutide is for people who are 18 years old and older with a Body Mass Index of 27 with one or more comorbidities like high blood pressure, high cholesterol, and diabetes type 2 that are caused by being overweight or obese or having a body mass index over 30.

In four 68-week studies, the safety and effectiveness of semaglutide 2.4 mg were examined. Three were randomized, double-blind, placebo-controlled studies with 16-week dosage increases, and one was a randomized, double-blind, placebo-controlled withdrawal study in which patients were allocated semaglutide continued therapy or switched to placebo.

In the STEP 1 research trial, published in The New England Journal of Medicine, people with obesity who received semaglutide 2.4 mg lost significantly more weight than those who received a placebo. More than half of the patients lost 15% of their body weight.

In JAMA-published follow-up results from the STEP 4 research, persons with obesity who maintained semaglutide 2.4 mg after 20 weeks showed sustained weight reduction or weight maintenance in comparison to those who changed to a placebo.

After randomization, the projected mean weight change from week 20 to week 68 with continuing semaglutide was –7.9% compared to a mean rise of 6.9% among patients moved to placebo (difference of –14.8 percentage points; 95% CI, –16 to –3.5). Compared to placebo, waist circumference and BMI reduced from week 20 to 68 (mean difference, –9.7 cm; 95% CI, –10.9 to –8.5) and from week 20 to 68, respectively (mean difference, –4.7; 95% CI, –5.2 to –4.3). During the maintenance phase, systolic blood pressure remained steady for those allocated semaglutide but rose for those assigned placebo (difference, –3.9 mm Hg; 95% CI, –5.9 to –2). Physical function ratings improved in the semaglutide group as compared to the placebo group.

According to the FDA, the dose of semaglutide must be raised gradually over 16 to 20 weeks to 2.4 mg once weekly in order to prevent gastrointestinal adverse effects.

Do not use Semaglutide 2.4 mg with other GLP-1 receptor agonists, medications – products containing semaglutide or other supplements, or medications for weight loss. This includes prescription medications, herbal remedies, and over-the-counter products unless approved by your physician.

Semaglutide 2.4 mg has not been assessed in patients with a history of pancreatitis.

Semaglutide should not be used in patients with a personal or family history of medullary thyroid carcinoma or in patients with multiple endocrine neoplasia syndrome type 2.
